Overview
Pucotenlimab + MRG002 is an investigational combination therapy for the treatment of HER2-expressing solid tumors, including HER2-positive cancers of unknown primary and urothelial carcinoma. Pucotenlimab is a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance anti-tumor immune responses. MRG002 is a novel antibody-drug conjugate (ADC) composed of a sugar-modified trastuzumab linked via a cleavable valine-citrulline linker to the cytotoxic payload monomethyl auristatin E (MMAE). The combination aims to leverage both immune checkpoint blockade and targeted cytotoxicity against HER2-expressing tumor cells. Clinical trials are ongoing in Phase II for various solid tumors, with encouraging preliminary efficacy and safety data reported in urothelial carcinoma and other HER2-positive malignancies[1][4][5][7].
