This neoadjuvant chemo-immunotherapy regimen is a multimodal treatment approach being investigated by Tang-Du Hospital for the treatment of resectable locally advanced gastroesophageal junction (GEJ) adenocarcinoma. The regimen combines **pucotenlimab** (HX008), a humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor to restore anti-tumor immune responses, with a cytotoxic chemotherapy backbone. This backbone consists of **nab-paclitaxel** (an albumin-bound nanoparticle formulation of paclitaxel that stabilizes microtubules), **tegafur** (an oral fluoropyrimidine antimetabolite that inhibits thymidylate synthase), and **carboplatin** (a platinum-based agent that induces DNA alkylation and cross-linking). The combination is administered prior to surgery to reduce tumor volume, eliminate micrometastases, and improve the feasibility of radical resection in patients with cT2N+M0 or cT3-4bNxM0 GEJ tumors.
