Pucotenlimab + vorolanib is a combination therapy comprising a humanized IgG4 monoclonal antibody targeting the PD-1 receptor and a small molecule multi-kinase inhibitor. Pucotenlimab (HX008) acts as an immune checkpoint inhibitor by blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell mediated anti-tumor immunity. Vorolanib (CM082) is a tyrosine kinase inhibitor that selectively targets vascular endothelial growth factor receptors (VEGFR1, VEGFR2, VEGFR3), platelet-derived growth factor receptors (PDGFR), and c-Kit, effectively inhibiting tumor angiogenesis and cell proliferation. This combination is being investigated as a neoadjuvant treatment for patients with high-risk localized clear cell renal cell carcinoma (ccRCC) to reduce tumor complexity and facilitate nephron-sparing surgery (partial nephrectomy).
