Drug intelligence / Profile preview
Purinostat mesylate + tislelizumab is a combination therapy under clinical investigation for hematologic malignancies and solid tumors. Purinostat mesylate is a highly potent and selective small-molecule inhibitor of histone deacetylase (HDAC), specifically targeting Class I (HDAC1, 2, 3, 8) and Class IIb (HDAC6, 10) isoforms, leading to tumor cell apoptosis through epigenetic modulation and enhancing antitumor immunity by remodeling the tumor microenvironment[1][2][3][4]. Tislelizumab is a humanized IgG4 monoclonal antibody that blocks the programmed cell death protein 1 (PD-1) immune checkpoint, thereby promoting T-cell-mediated antitumor responses. The combination is being explored to leverage purinostat mesylate’s dual cytotoxic and immunomodulatory properties with tislelizumab’s immune checkpoint inhibition. The combination aims to maximize tumor-killing efficacy and overcome resistance in relapsed/refractory lymphomas and advanced solid tumors[1].
