Overview

Pvs25-IMX313 + Matrix-M1 is a transmission-blocking vaccine candidate for malaria. It combines Pvs25-IMX313, a protein-nanoparticle vaccine based on the Plasmodium vivax transmission-blocking target antigen Pvs25 genetically fused to the IMX313 oligomerization domain, with Matrix-M1, a potent saponin-based adjuvant. The vaccine aims to induce an immune response that blocks the transmission of malaria parasites. Pvs25-IMX313 is expressed and secreted in the Pichia pastoris expression system, while Matrix-M1 comprises partially purified extracts of the Quillaja saponaria Molina tree bark, phosphatidylcholine, and cholesterol, formulated as a 40nm-sized complex. Matrix-M1 efficiently activates and recruits immune cells to the draining lymph node, including T-cells, B-cells, NK-cells, dendritic cells, and granulocytes.
