This is a combination chemotherapy regimen consisting of belinostat (PXD101), cisplatin, doxorubicin, and cyclophosphamide. Belinostat is a histone deacetylase (HDAC) inhibitor that induces transformed cell differentiation, cell cycle arrest, and apoptosis by affecting chromatin structure and gene expression. It is approved for the treatment of peripheral T-cell lymphoma and has been studied in combination with other chemotherapeutic agents for various solid tumors and hematological malignancies[3][4]. Cisplatin is a platinum-based chemotherapeutic agent that forms DNA crosslinks leading to apoptosis. Doxorubicin is an anthracycline antibiotic that intercalates DNA strands and inhibits topoisomerase II activity. Cyclophosphamide is an alkylating agent used in the treatment of various cancers; it works by slowing or stopping the growth of cancer cells through DNA crosslinking[6][8][10]. This multi-agent regimen aims to leverage synergistic antitumor effects from different mechanisms.
