Overview

Pyrimethamine + amodiaquine is a fixed-dose combination of two antimalarial agents used primarily for the treatment and prevention of malaria. Amodiaquine is a synthetic 4-aminoquinoline with schizonticidal activity against *Plasmodium falciparum*, *P. vivax*, and *P. ovale* by destroying intraerythrocytic forms; its mechanism involves inhibition of heme polymerase in the parasite, leading to toxic accumulation of free heme and parasite death[3][5]. Pyrimethamine is a diaminopyrimidine that acts as an antifolate by inhibiting dihydrofolate reductase in plasmodia, thereby blocking DNA synthesis and cell multiplication during schizont formation[5][8]. The combination exploits different mechanisms to enhance efficacy against malaria parasites and reduce resistance development. It is mainly indicated for curative treatment or intermittent preventive therapy (IPT) in malaria-endemic regions[2][5].
