A combination therapy of pyrotinib and thalidomide being investigated for the treatment of advanced non-small-cell lung cancer (NSCLC) patients with human epidermal growth factor receptor 2 (HER2) exon 20 insertions. Pyrotinib is an irreversible EGFR/HER2 dual tyrosine kinase inhibitor, while thalidomide is an antiangiogenic and immunomodulatory drug that functions as a cereblon-based molecular glue. The addition of thalidomide to pyrotinib is expected to both increase clinical benefit and reduce the incidence of pyrotinib-related diarrhea.
