Overview

PYTX-004 + anti-PD-1 is a preclinical-stage combination therapy being developed by Papyrus Therapeutics for the treatment of solid tumors, with a particular focus on ovarian cancer. The combination consists of PYTX-004, a recombinant Fc-fusion protein (OPCML-Fc) designed to restore the function of the Opioid Binding Protein/Cell Adhesion Molecule-like (OPCML) tumor suppressor, and an anti-PD-1 checkpoint inhibitor. OPCML is a cell-surface protein that is frequently epigenetically silenced in various cancers; when restored, it acts as a broadly acting tumor suppressor by binding and inactivating nine activated receptor tyrosine kinases (RTKs), including HER2, HER4, FGFR1, FGFR3, EphA2, AXL, TYRO3, MET, and VEGFR3. By combining this multi-RTK inhibition and direct oncolysis with the immune-stimulating effects of PD-1 blockade, the therapy aims to achieve synergistic anti-tumor activity, suppress angiogenesis, and activate robust anti-tumor immunity.
