PYY3-36 + oxyntomodulin is a combination of two endogenous gut hormones—peptide YY (PYY) 3–36 and oxyntomodulin—administered together to reduce appetite and promote weight loss. Both peptides are secreted by intestinal L-cells after meals and act as satiety signals. PYY3-36 is a selective agonist of the Y2 receptor, while oxyntomodulin acts as an agonist at both the glucagon-like peptide 1 (GLP-1) receptor and the glucagon receptor, though its primary anorectic effect is mediated via GLP-1 receptor activation. Clinical studies have shown that coadministration of these hormones produces additive reductions in energy intake compared to either hormone alone, suggesting potential utility for obesity treatment[1][10]. The combination has also been investigated for effects on glucose metabolism in prediabetes and type 2 diabetes[2][7]. This approach mimics postprandial hormone profiles seen after bariatric surgery.
