QLS1304 + fulvestrant + QLC1401 is a combination therapy regimen being developed by Qilu Pharmaceutical for the treatment of estrogen receptor-positive (ER+), human epidermal growth factor receptor 2-negative (HER2-) breast cancer. The combination consists of QLS1304, a novel oral small-molecule inhibitor of lysine acetyltransferases 6A and 6B (KAT6A/B); fulvestrant, an intramuscularly administered selective estrogen receptor degrader (SERD); and QLC1401, an investigational oral SERD. QLS1304 targets the 8p11.2 genomic amplification, a common alteration in breast cancer that leads to KAT6A overexpression, which in turn promotes ER-mediated transcription and cell cycle progression. By combining KAT6A/B inhibition with endocrine therapies like fulvestrant or QLC1401, the regimen aims to synergistically suppress tumor growth and overcome resistance to standard endocrine treatments. This combination is currently being evaluated in Phase Ib/II clinical trials for patients with advanced or metastatic breast cancer who have progressed on prior endocrine-based therapies.
