QTX-2101 + all-trans retinoic acid (ATRA) is an investigational combination therapy being developed by Quetzal Therapeutics for the treatment of newly diagnosed low- or intermediate-risk acute promyelocytic leukemia (APL). QTX-2101 is an oral capsule formulation of arsenic trioxide (ATO), designed to provide a more convenient alternative to the standard-of-care intravenous ATO (Trisenox). The mechanism of action involves a synergistic approach: arsenic trioxide induces the degradation of the PML-RARA fusion protein, the primary driver of APL, while ATRA (tretinoin) acts as a differentiating agent that promotes the maturation of promyelocytes into mature granulocytes. This combination aims to maintain the high efficacy of the ATO-ATRA regimen while significantly reducing the treatment burden associated with frequent clinic visits and lengthy intravenous infusions. The therapy is currently being evaluated in a global Phase III clinical trial and has received FDA Fast Track designation.
