The quadrivalent hand, foot and mouth disease (HFMD) vaccine is a multi-valent immunization designed to protect against the four primary enteroviruses responsible for HFMD outbreaks: Enterovirus A71 (EV71), Coxsackievirus A16 (CA16), Coxsackievirus A6 (CA6), and Coxsackievirus A10 (CA10). While monovalent EV71 vaccines have been successful in reducing severe cases of HFMD, they offer limited cross-protection against other coxsackieviruses, which have increasingly become dominant strains in various regions. Leading candidates, such as SinoCellTech's SCT-V522, utilize recombinant virus-like particle (VLP) technology to present the structural VP1 proteins of all four serotypes, while other programs, such as those by Sinopharm/CNBG, utilize inactivated virus platforms. These vaccines aim to elicit a robust neutralizing antibody response to prevent both mild and severe clinical manifestations of the disease in pediatric populations.
