This investigational quadrivalent recombinant influenza vaccine (RIV4) is being developed by Sanofi Pasteur using a baculovirus expression system. Unlike traditional egg-based vaccines, this platform produces recombinant hemagglutinin (rHA) proteins in insect cells, which allows for a faster manufacturing process and avoids egg-adaptive mutations that can reduce vaccine effectiveness. The specific formulation evaluated in Phase 1 trials (such as NCT03677141) explores the use of alternative H3 hemagglutinin antigen variants and the addition of an adjuvant to enhance immunogenicity and broaden antigenic coverage against drifting H3N2 influenza strains. The vaccine is designed to induce active immunization against four influenza strains: two influenza A strains (H1N1 and H3N2) and two influenza B strains (Victoria and Yamagata lineages).
