AB680 + ABI-007 is a combination therapy consisting of **quemliclustat** (AB680), a potent and selective small molecule inhibitor of **CD73** (ecto-5'-nucleotidase), and **nab-paclitaxel** (ABI-007), a nanoparticle albumin-bound formulation of the microtubule inhibitor paclitaxel. CD73 is an enzyme that converts extracellular adenosine monophosphate (AMP) into adenosine, a potent immunosuppressive metabolite in the tumor microenvironment. By inhibiting CD73, quemliclustat aims to restore anti-tumor immune responses and enhance the efficacy of chemotherapy and immunotherapy. Nab-paclitaxel provides cytotoxic activity by stabilizing microtubules, leading to cell cycle arrest and apoptosis. This combination is being developed by **Taiho Pharmaceutical** in collaboration with Arcus Biosciences, primarily for the treatment of **pancreatic ductal adenocarcinoma (PDAC)**, where it is often evaluated as part of a regimen including gemcitabine and the anti-PD-1 antibody zimberelimab.
