R-(-)-BPAP (R-(-)-1-(benzofuran-2-yl)-2-propylaminopentane) is a synthetic small molecule that acts as a catecholaminergic and serotonergic activity enhancer (CAE/SAE). Developed by Fujimoto Pharmaceutical, it represents a unique pharmacological class that selectively potentiates the impulse-evoked release of catecholamines (dopamine and norepinephrine) and serotonin in the brain. Unlike traditional stimulants or monoamine oxidase inhibitors, R-(-)-BPAP does not trigger massive, non-vesicular release of neurotransmitters or inhibit their metabolism at therapeutic doses, potentially offering a more refined modulation of monoaminergic pathways. Beyond its effects on neurotransmitter release, R-(-)-BPAP has demonstrated neuroprotective properties in preclinical models, including the stabilization of mitochondrial membrane potential and the induction of anti-apoptotic factors such as Bcl-2 and neurotrophic factors like BDNF and GDNF. It has been primarily investigated for the treatment of Parkinson's disease, Alzheimer's disease, and clinical depression.
