Overview

R1507 + bevacizumab + paclitaxel is an investigational combination regimen comprising three agents: - **R1507** is a fully human IgG1 monoclonal antibody that specifically targets the insulin-like growth factor 1 receptor (IGF-1R), inhibiting IGF-1- and IGF-2-mediated signaling and suppressing anchorage-independent growth of malignant cells. - **Bevacizumab** is a recombinant humanized monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis, the process by which tumors develop new blood vessels. - **Paclitaxel** is a chemotherapeutic agent classified as a taxane; it acts by stabilizing microtubules and preventing cell division, ultimately causing cancer cell death. This combination has been studied in a phase 1b setting, notably as part of a regimen for patients with various advanced cancers, including non-small cell lung cancer and nasopharyngeal cancer[1]. The rationale for the combination is to simultaneously block tumor growth and spread via IGF-1R, inhibit tumor blood supply with anti-VEGF therapy, and induce cancer cell death with cytotoxic chemotherapy.
