R1507 + erlotinib is a combination therapy consisting of two agents: - **R1507** is a fully human recombinant monoclonal antibody (IgG1 subclass) that selectively targets and inhibits the insulin-like growth factor-1 receptor (IGF-1R), blocking its activation and downstream signaling. - **Erlotinib** is an orally administered small molecule inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase. This combination was investigated for the treatment of advanced-stage non-small cell lung cancer (NSCLC), based on preclinical evidence suggesting that dual inhibition of IGF-1R and EGFR could overcome resistance to EGFR inhibitors. In clinical trials, patients received either weekly or every-three-week intravenous infusions of R1507 in addition to daily oral erlotinib. However, phase II studies showed no significant improvement in progression-free survival or overall survival compared to erlotinib alone in unselected NSCLC populations[1][2][3][6]. The development of this combination has not progressed further due to lack of efficacy benefit.
