Rabbit anti-thymocyte globulin is a polyclonal antibody preparation derived from rabbits immunized with human thymocytes. It is used as an immunosuppressive agent to prevent and treat acute rejection in organ transplantation, particularly kidney and liver transplants. The mechanism of action involves binding to multiple T-cell specific antigens, leading to T-cell depletion via complement-mediated cytotoxicity or apoptosis, and induction of regulatory T-cells[1][3][4][5][6]. Cyclosporine and tacrolimus are calcineurin inhibitors that suppress the immune response by inhibiting interleukin 2 production in activated T-helper cells, thereby reducing proliferation of cytotoxic T-cells responsible for graft rejection[8]. This combination regimen is commonly used for prophylaxis against acute rejection in solid organ transplantation.
