Overview

**rAd-p53 + cisplatin + paclitaxel** is a combination therapy used in the treatment of certain cancers, most notably cervical cancer. - **rAd-p53** is a gene therapy product consisting of a replication-defective recombinant human adenovirus vector carrying the wild-type p53 tumor suppressor gene. It restores p53 function in tumor cells, leading to cell cycle arrest, induction of programmed cell death (apoptosis), inhibition of angiogenesis via VEGF suppression, and enhanced sensitivity to chemotherapy and radiotherapy[1][5][3]. - **Cisplatin** is a small molecule platinum-based chemotherapy that forms DNA crosslinks, disrupting DNA replication and leading to apoptosis. - **Paclitaxel** is a taxane-class microtubule stabilizing chemotherapy that interferes with mitosis, leading to mitotic arrest and cell death[2][4][6]. This triple combination leverages gene therapy to reactivate defective tumor suppressor pathways while combining standard cytotoxic chemotherapies to maximize anticancer efficacy. This regimen is most commonly studied and used for locally advanced cervical cancer, but individual components are used in various tumor types.
