This combination treatment regimen is being evaluated in a Phase II clinical trial (NCT05493306) for the treatment of HER2-negative advanced breast cancer with brain metastases. The regimen consists of local radiotherapy (either stereotactic radiotherapy or whole-brain radiotherapy) combined with anlotinib, an oral multi-target tyrosine kinase inhibitor, and a microtubule inhibitor (such as eribulin or taxanes). Anlotinib targets several pathways involved in tumor angiogenesis and growth, including VEGFR, FGFR, PDGFR, and c-Kit. The microtubule inhibitor provides cytotoxic effects by disrupting tubulin polymerization or depolymerization, while radiotherapy provides local control of brain lesions. This multi-modal approach aims to improve intracranial and systemic disease control in a high-need patient population.
