rAdp53 + cisplatin is a combination therapy consisting of recombinant adenovirus-p53 (rAdp53), a gene therapy delivering the wild-type p53 tumor suppressor gene via an adenoviral vector, and cisplatin, a platinum-based chemotherapy agent. rAdp53 works by restoring or enhancing p53 function in tumor cells with dysfunctional or mutated p53 genes, leading to cell cycle arrest and apoptosis. Cisplatin acts as an alkylating agent that cross-links DNA strands and disrupts DNA replication and transcription, resulting in cancer cell death. This combination has been studied for enhanced efficacy in various cancers including head and neck cancer, cervical cancer, lung cancer, liver cancer, colorectal cancer, ovarian cancer, nasopharyngeal carcinoma and malignant pleural effusion. Clinical studies have shown improved response rates and survival outcomes when rAdp53 is combined with chemotherapy such as cisplatin compared to chemotherapy alone[6][8].
