Overview

Raltitrexed + oxaliplatin (also known as the RALOX regimen) is a combination chemotherapy regimen being investigated for the treatment of advanced malignant biliary system tumors. Raltitrexed is a quinazoline folate analogue that functions as a potent and specific inhibitor of thymidylate synthase (TS), an enzyme essential for DNA synthesis. Oxaliplatin is a third-generation platinum-based antineoplastic agent that works by forming DNA adducts, which lead to intra-strand and inter-strand cross-links, effectively inhibiting DNA replication and transcription. This combination is primarily being studied as a second-line therapy in patients with unresectable advanced cholangiocarcinoma or gallbladder carcinoma who have failed first-line treatments.
