**Raludotatug deruxtecan + paclitaxel** is an investigational drug combination comprising raludotatug deruxtecan—a first-in-class CDH6-directed antibody-drug conjugate (ADC) using Daiichi Sankyo's DXd ADC technology—and paclitaxel, a well-established cytotoxic chemotherapeutic agent. Raludotatug deruxtecan consists of a humanized anti-CDH6 IgG1 monoclonal antibody linked to a topoisomerase I inhibitor payload (an exatecan derivative, DXd) via a tetrapeptide-based cleavable linker. Paclitaxel inhibits microtubule disassembly during cell division, leading to cell cycle arrest and apoptosis. The combination is under investigation for patients with platinum-resistant ovarian cancer and related gynecological tumors, including those previously treated with platinum-based chemotherapy. Development is led by Daiichi Sankyo and Merck, with clinical evaluation in phase 1b/2 trials for relapsed high-grade serous ovarian cancer and related indications[1][2][3][7][9].
