Ralzapastotug (AB308) + zimberelimab is a combination immunotherapy regimen being investigated for the treatment of advanced solid tumors and hematologic malignancies. Ralzapastotug is an Fc-enabled anti-TIGIT humanized IgG1 monoclonal antibody that blocks the TIGIT-CD155 interaction, reversing TIGIT-mediated T cell inhibition and enabling CD226-mediated signaling to activate the immune system against cancer cells. It also has the capacity to induce FcγR-mediated signaling and enable NK cell-mediated antibody-dependent cell-mediated cytotoxicity (ADCC) against TIGIT-expressing target cells. Zimberelimab is an anti-PD-1 monoclonal antibody. The combination aims to enhance immune responses against cancer by targeting two distinct immune checkpoint pathways.
