This is a combination of two pharmaceutical drugs, ramelteon and zolpidem, with placebo as a control. Ramelteon is a melatonin receptor agonist that selectively binds to MT1 and MT2 receptors in the suprachiasmatic nucleus of the brain, mimicking the action of endogenous melatonin to regulate circadian rhythms and promote sleep onset. Zolpidem is a non-benzodiazepine hypnotic that acts as an agonist at the GABA-A receptor complex, enhancing inhibitory neurotransmission to induce sedation and facilitate both sleep initiation and maintenance. Placebo serves as an inactive comparator in clinical trials. This combination has been studied in randomized controlled trials for their effects on sleep parameters such as total sleep time, latency, REM duration, body sway (as a measure of residual effect), and safety profiles[2]. The use of both active drugs together may increase central nervous system depressant effects; co-administration is generally not recommended outside research settings due to potential additive or synergistic sedative effects[1][5].
