ramucirumab + merestinib

Ramucirumab + merestinib is a combination regimen comprising two investigational anticancer agents: ramucirumab, a monoclonal antibody targeting vascular endothelial growth factor receptor 2 (VEGFR2), and merestinib, an oral type II MET kinase inhibitor that also inhibits other receptor tyrosine kinases. The combination was evaluated primarily for patients with metastatic colorectal cancer (mCRC) and in advanced or metastatic biliary tract cancer. The mechanism is to cotarget angiogenesis (via VEGFR2 blockade) and tumor cell growth/survival pathways (via MET and related kinases inhibition). Preliminary data suggest the combination is generally well tolerated and may provide clinical benefit in selected, heavily pretreated mCRC patients, though robust clinical efficacy has not been established in randomized prospective trials[1][4][5]. In advanced biliary tract cancer, the combination has not been reported as a formal arm; ramucirumab and merestinib were tested as separate additions to first-line chemotherapy, each showing tolerable safety profiles but no survival benefit[2][5][6].
