Overview

**Ravulizumab + rHuPH20** is a combination of ravulizumab, a long-acting monoclonal antibody inhibitor of the complement protein C5, and rHuPH20, a recombinant human hyaluronidase PH20. Ravulizumab inhibits the terminal complement pathway by binding to C5, thereby preventing its cleavage into C5a and C5b and blocking formation of the membrane attack complex (MAC), effectively reducing complement-mediated inflammation and cell lysis. rHuPH20 acts by transiently depolymerizing hyaluronan in the subcutaneous (SC) extracellular matrix, enhancing bulk fluid flow, and improving the dispersion and absorption of therapeutics administered subcutaneously. Co-administration (simultaneous or sequential) or co-formulation of these agents enables subcutaneous delivery of ravulizumab, overcoming barriers of IV administration for conditions such as paroxysmal nocturnal hemoglobinuria (PNH) and atypical hemolytic uremic syndrome (aHUS)[1][2][3][5][6][7].
