RC88 + sintilimab is an investigational combination therapy composed of RC88, a novel **mesothelin (MSLN)-targeting antibody-drug conjugate (ADC)**, and sintilimab, a **monoclonal antibody immune checkpoint inhibitor targeting PD-1**. RC88 is developed by RemeGen and comprises a recombinant humanized anti-MSLN monoclonal antibody conjugated to the microtubule inhibitor MMAE via a cleavable linker. It selectively targets MSLN-expressing tumor cells, delivers the cytotoxic payload to induce cell cycle arrest and apoptosis, and is designed to provoke immunogenic cell death. Sintilimab, marketed as TYVYT, is co-developed by Innovent and Eli Lilly, and binds to PD-1, blocking the PD-1/PD-L1 interaction to reactivate T cells against tumor cells. Preclinical data support a synergistic anti-tumor effect, where RC88 enhances immunogenic cell death and antigen presentation, thus improving the efficacy of PD-1 blockade via Sintilimab. The combination is currently under clinical investigation for advanced solid tumors, focusing on patient populations with high unmet clinical needs such as platinum-resistant gynecologic malignancies and other MSLN-positive cancers[1][2][3][4][5][6][7].
