Overview

rDEN3-3'D4delta30 is a live-attenuated dengue virus vaccine candidate specifically targeting serotype 3 (DENV-3). Developed by the National Institute of Allergy and Infectious Diseases (NIAID), it was designed to overcome the insufficient attenuation observed in earlier DENV-3 candidates (such as rDEN3delta30). The vaccine is a chimeric virus consisting of the wild-type DENV-3 (strain Sleman/78) genetic background, but with its 3' untranslated region (UTR) replaced by the 3' UTR of the attenuated rDEN4delta30 vaccine candidate. This modification incorporates a 30-nucleotide deletion (delta30) that provides the necessary attenuation while maintaining immunogenicity. It has been evaluated in Phase 1 clinical trials as a monovalent vaccine and serves as the DENV-3 component in the tetravalent dengue vaccine formulations TV003 and TV005.
