recMAGE-A3 + AS15 is an investigational cancer immunotherapy composed of a recombinant MAGE-A3 protein (a tumor-associated antigen) combined with the AS15 immunostimulant adjuvant system. The therapy is designed to induce an immune response specifically against tumors expressing the MAGE-A3 antigen, which is present in various cancers such as melanoma and non-small cell lung cancer (NSCLC). The mechanism of action involves stimulating both humoral and cellular immunity, particularly enhancing CD4+ T cell responses against MAGE-A3-expressing tumor cells. The adjuvant AS15 contains QS-21 Stimulon, monophosphoryl lipid A, and CpG7909 (a TLR9 agonist) in a liposomal formulation to boost the immune response. Developed primarily by GSK, this combination has been evaluated in multiple phase II and III clinical trials for melanoma, NSCLC, and multiple myeloma[2][4][6][7][8].
