Overview

A combination vaccine composed of a **recombinant hemagglutinin (rHA) protein derived from influenza A H5N1** and **glucopyranosyl lipid A (GLA)**, typically formulated in a stable emulsion (GLA-SE). The rHA component stimulates production of neutralizing antibodies against the influenza H5N1 virus by mimicking the virus’s major surface glycoprotein, hemagglutinin, which is essential for viral entry into host cells. GLA acts as a potent adjuvant; as a synthetic Toll-like receptor 4 (TLR4) agonist, it enhances the vaccine-induced immune response by promoting Th1-biased (cellular and humoral) immunity. This formulation is being developed primarily as a preventive vaccine for pandemic or avian influenza H5N1, aiming to provide broader and more robust immunogenicity compared to non-adjuvanted or alum-adjuvanted H5N1 vaccines. GLA-SE significantly increases immunogenicity and enables dose sparing relative to non-adjuvanted formulations[5].
