recombinant human endostatin adenovirus + PD-1 inhibitor is an experimental combination therapy being developed by Sichuan University for the treatment of recurrent or metastatic head and neck cancer and esophageal squamous cell carcinoma (ESCC). The regimen combines an intratumorally administered gene therapy, consisting of a replication-deficient adenovirus vector encoding the human endostatin gene (Ad-endostatin), with an intravenously administered PD-1 inhibitor (such as pembrolizumab or camrelizumab). Ad-endostatin is designed to induce local, sustained expression of endostatin within the tumor microenvironment, where it acts as a potent anti-angiogenic agent by inhibiting endothelial cell proliferation and normalizing tumor vasculature. The PD-1 inhibitor blocks the interaction between the PD-1 receptor on T cells and its ligands, thereby reversing immune suppression. This dual approach aims to enhance anti-tumor efficacy by combining the vascular-normalizing effects of endostatin with the immune-activating effects of checkpoint blockade, potentially improving T-cell infiltration and overall therapeutic response.
