This therapeutic regimen is a triple combination therapy consisting of toripalimab, recombinant human interferon-α1b (IFN-α1b), and anlotinib hydrochloride, primarily investigated for the treatment of advanced unresectable melanoma. Toripalimab is a humanized IgG4 monoclonal antibody that functions as a checkpoint inhibitor by targeting the programmed cell death-1 (PD-1) receptor, thereby enhancing the immune system's ability to attack cancer cells. Anlotinib hydrochloride is a multi-targeted tyrosine kinase inhibitor (TKI) that inhibits angiogenesis and tumor proliferation by targeting vascular endothelial growth factor receptors (VEGFR), fibroblast growth factor receptors (FGFR), platelet-derived growth factor receptors (PDGFR), and c-kit. Recombinant human interferon-α1b provides additional immunomodulatory and antiproliferative effects. This combination is being evaluated in clinical trials led by Xijing Hospital to improve response rates in patients with advanced melanoma, particularly those with mucosal or acral subtypes common in Chinese populations.
