recombinant interleukin-12 + recombinant interferon alfa is an investigational combination immunotherapy regimen consisting of two recombinant cytokines. Recombinant interleukin-12 (IL-12) is a heterodimeric cytokine that acts as a potent inducer of interferon-gamma production and a key regulator of Th1-type immune responses, activating both natural killer (NK) cells and cytotoxic T lymphocytes (CTLs). Recombinant interferon alfa (IFN-α) is a type I interferon that exerts direct anti-proliferative and pro-apoptotic effects on malignant cells while enhancing the expression of MHC class I antigens to facilitate tumor recognition by the immune system. This specific regimen, studied by the National Cancer Institute (NCI), involves the sequential administration of IL-12 followed by IFN-α to evaluate safety and potential synergistic anti-tumor activity in patients with advanced solid tumors, including malignant melanoma, who have failed standard therapies.
