Recombinant PRAME protein + AS15 adjuvant system (GSK2302032A) is an antigen-specific cancer immunotherapeutic (ASCI) developed by GSK. It consists of a recombinant form of the human PRAME (Preferentially Expressed Antigen of Melanoma) protein — a tumor-associated antigen frequently overexpressed across multiple tumor types — combined with GSK's proprietary AS15 adjuvant system. The AS15 adjuvant is a potent liposomal formulation containing CpG 7909 (a TLR9 agonist), monophosphoryl lipid A (MPL, a TLR4 agonist), and QS-21 (a saponin). Upon intramuscular administration, the vaccine is designed to stimulate the host immune system to mount a cytotoxic T-lymphocyte (CTL) response against PRAME-overexpressing tumor cells, resulting in tumor cell lysis. The drug was primarily evaluated in Phase I and Phase II clinical trials as an adjuvant therapy in patients with resected non-small cell lung cancer (NSCLC) and metastatic melanoma. Clinical development was halted in 2016 following negative results with a similar ASCI (recMAGE-A3+AS15) in NSCLC patients.
