Refnot is a recombinant fusion protein consisting of tumor necrosis factor-alpha (TNF-α) and thymosin alpha-1 (Tα1), joined by an Ile-Asp-Met tripeptide linker. Developed by Refnot-Pharm Ltd, the drug is designed to leverage the potent antitumor cytotoxic activity of TNF-α while utilizing the immunomodulatory properties of thymosin alpha-1 to enhance T-cell function and reduce the systemic toxicity typically associated with native TNF. This hybrid biologic exhibits a toxic potential more than 100 times lower than recombinant TNF alone, allowing for safer therapeutic administration. In clinical settings, Refnot is evaluated both as a monotherapy and in combination with various chemotherapy regimens for the treatment of disseminated solid tumors, including melanoma, breast cancer, and gastrointestinal malignancies.
