REGN17372 + linvoseltamab is an investigational combination therapy being developed by Regeneron Pharmaceuticals for the treatment of relapsed or refractory multiple myeloma. This combination utilizes two distinct bispecific antibodies: linvoseltamab (REGN5458), a B-cell maturation antigen (BCMA) x CD3 T-cell engager, and REGN17372, a G-protein coupled receptor class C group 5 member D (GPRC5D) x CD28 costimulatory bispecific antibody. While linvoseltamab provides the primary T-cell activation signal by bridging BCMA-expressing myeloma cells with CD3 on T cells, REGN17372 provides a secondary costimulatory signal by bridging GPRC5D-expressing myeloma cells with CD28 on T cells. This dual-targeting approach is designed to enhance T-cell activation, proliferation, and cytotoxicity while potentially overcoming resistance mechanisms associated with single-agent T-cell engagers.
