REGN5093-M114 + cemiplimab is an investigational combination therapy of two agents: REGN5093-M114, an antibody-drug conjugate (ADC) targeting MET (mesenchymal epithelial transition factor), and cemiplimab, an immune checkpoint inhibitor targeting PD-1. REGN5093-M114 consists of a biparatopic METxMET human IgG4p bispecific antibody conjugated via a novel linker to the cytotoxic payload M24 (a maytansine derivative inhibiting microtubule assembly) and is designed to selectively kill MET-overexpressing cancer cells by delivering the cytotoxic payload directly into these cells. Cemiplimab is a fully human monoclonal antibody that blocks PD-1, enabling anti-tumor immune responses. The combination aims to address advanced non-small cell lung cancer (NSCLC) characterized by MET overexpression, including cases resistant to EGFR tyrosine kinase inhibitors. The regimen is under evaluation for safety, tolerability, pharmacokinetics, and antitumor activity in ongoing phase 1/2 trials[1][2][3][5][6][7].
