Overview

Regorafenib + camrelizumab is a combination therapy under investigation for the treatment of various cancers, including metastatic colorectal cancer and hepatocellular carcinoma. Regorafenib is an orally administered small molecule inhibitor that targets multiple membrane-bound and intracellular kinases involved in tumor angiogenesis, oncogenesis, metastasis, and maintenance of the tumor microenvironment. Its primary targets include VEGFR1/2/3, KIT, RET, PDGFR-alpha/beta, FGFR1/2, TIE2, RAF1/BRAF/BRAFV600E among others[3][6][8]. Camrelizumab is a humanized monoclonal antibody that acts as an immune checkpoint inhibitor by targeting programmed cell death protein 1 (PD-1), thereby enhancing anti-tumor immune responses[1]. The combination aims to leverage both direct anti-tumoral effects from kinase inhibition and immune-mediated tumor destruction via checkpoint blockade. This regimen has shown feasibility with manageable toxicity in early studies for metastatic colorectal cancer with microsatellite stability[1], and it is being explored in other solid tumors.
