Overview

Regorafenib + envafolimab is an investigational combination therapy consisting of regorafenib, a multi-kinase inhibitor, and envafolimab, a PD-L1-targeting monoclonal antibody. Regorafenib inhibits multiple protein kinases involved in tumor angiogenesis (such as VEGFR), oncogenesis (such as KIT, RET), and the tumor microenvironment. Envafolimab is a humanized single-domain anti-PD-L1 antibody designed to block the interaction between PD-L1 and its receptor PD-1 on T cells, thereby enhancing antitumor immune responses. This combination aims to leverage both targeted inhibition of cancer cell signaling pathways and immune checkpoint blockade for synergistic antitumor activity. The primary indications under investigation are advanced or metastatic solid tumors such as colorectal cancer and gastric cancer.
