Relacorilant + enzalutamide is an investigational combination therapy comprising the selective glucocorticoid receptor modulator (SGRM) relacorilant and the androgen receptor (AR) inhibitor enzalutamide. Developed by Corcept Therapeutics in collaboration with the University of Chicago, this combination is primarily being evaluated for the treatment of metastatic castration-resistant prostate cancer (mCRPC). The therapeutic rationale is based on the observation that glucocorticoid receptor (GR) signaling can compensate for inhibited AR signaling, providing a survival pathway for cancer cells and contributing to enzalutamide resistance. By concurrently inhibiting both the GR and AR, the combination aims to block these escape mechanisms and enhance anti-tumor activity.
