Overview

This combination immunotherapy regimen is being investigated in a Phase 1/2 clinical trial for the treatment of advanced ovarian, fallopian tube, and primary peritoneal cancers. The therapy consists of autologous tumor-infiltrating lymphocytes (TILs) administered following lymphodepleting chemotherapy, combined with three immune checkpoint inhibitors: relatlimab, nivolumab, and ipilimumab. Relatlimab is a monoclonal antibody that targets Lymphocyte Activation Gene-3 (LAG-3), an inhibitory receptor often expressed on T-cells that limits their anti-tumor activity. Nivolumab and ipilimumab target the PD-1 and CTLA-4 pathways, respectively. By blocking these three distinct immune checkpoints, the regimen aims to prevent T-cell exhaustion and enhance the efficacy of the infused TILs within the immunosuppressive tumor microenvironment of gynecological malignancies.
