**REOLYSIN + gemcitabine** is a combination therapy composed of REOLYSIN (a proprietary formulation of wild-type human reovirus serotype 3, Dearing strain), an oncolytic virus, combined with gemcitabine, a nucleoside analog chemotherapeutic. REOLYSIN selectively infects and lyses cancer cells with activated RAS pathways, leading to immunogenic tumor cell death, apoptosis via endoplasmic reticulum stress, and immune activation. Gemcitabine interferes with DNA synthesis, inhibiting cell proliferation. The combination is being investigated for synergistic antitumor effects, particularly in cancers with high prevalence of RAS pathway mutations such as pancreatic adenocarcinoma. Primary indications studied include advanced and metastatic pancreatic cancer and other refractory solid tumors[1][2][3][4][5].
