Replicating RNA + LION is a vaccine platform developed by HDT Bio that combines self-amplifying replicon RNA (repRNA) with a Lipid InOrganic Nanoparticle (LION) delivery system. The repRNA is derived from the Venezuelan equine encephalitis virus (VEEV) TC-83 strain and encodes a replicase complex that enables intracellular amplification of the RNA, allowing for significantly lower doses compared to conventional mRNA vaccines. The LION delivery system is a stable, squalene-based oil-in-water emulsion that utilizes the cationic lipid DOTAP to electrostatically bind RNA to its surface, rather than encapsulating it. This configuration enhances stability, potentially allowing for room-temperature storage and distribution. The platform's lead candidate, HGC019 (GEMCOVAC-19), received emergency use authorization in India for COVID-19, and the technology is being explored for other infectious diseases including malaria and various viral pathogens.
