Drug intelligence / Profile preview
EIK1001 + Keytruda is an investigational combination immunotherapy regimen consisting of EIK1001 (resiquimod sulfate), a systemically administered dual agonist of Toll-like receptors 7 and 8 (TLR7/8), and pembrolizumab (Keytruda), an FDA-approved anti-PD-1 monoclonal antibody. Originally developed by Seven and Eight Biopharmaceuticals and subsequently licensed to Eikon Therapeutics, EIK1001 is designed to stimulate both innate and adaptive immune responses by activating myeloid and plasmacytoid dendritic cells, thereby enhancing antigen presentation and cytokine release. When combined with pembrolizumab, which blocks the immunosuppressive PD-1/PD-L1 pathway, the regimen provides a complementary, synergistic mechanism to overcome immune resistance and enhance T-cell-mediated tumor destruction. The combination is currently being evaluated in Phase 2 and Phase 2/3 clinical trials (such as the TeLuRide-005, TeLuRide-006, and TeLuRide-008 studies) for the treatment of advanced solid tumors, including first-line stage 4 non-small cell lung cancer (NSCLC) and advanced cutaneous melanoma.
