RSV-E (also referred to as RSV-E PO) is an oral solid dispersion formulation consisting of the natural polyphenol resveratrol (RSV) and the cationic copolymer Eudragit E PO. Developed to overcome the poor aqueous solubility and low oral bioavailability of raw resveratrol, this delivery system utilizes a 1:7 drug-to-polymer ratio to maintain resveratrol in an amorphous state. In preclinical models of rheumatoid arthritis, the formulation demonstrated enhanced oral bioavailability (doubling that of unformulated resveratrol) and significant anti-arthritic effects. These protective effects are primarily mediated through the modulation of key inflammatory and oxidative stress pathways, including the reduction of pro-inflammatory cytokines (IL-6, TNF-α, IL-1β) and malondialdehyde (MDA) levels, coupled with an increase in the anti-inflammatory cytokine IL-10 and the antioxidant enzyme superoxide dismutase (SOD).
