Overview

**Retifanlimab + carboplatin + nab-paclitaxel** is a three-drug combination regimen used as first-line therapy for advanced or metastatic squamous cell carcinoma of the anal canal (SCAC). Retifanlimab is a humanized monoclonal antibody that targets programmed cell death protein-1 (PD-1), functioning as an immune checkpoint inhibitor[1][3]. Carboplatin is a platinum-based chemotherapeutic agent that forms DNA crosslinks causing apoptosis. Nab-paclitaxel (albumin-bound paclitaxel) is a microtubule inhibitor delivered by albumin nanoparticles, enabling increased intratumoral concentrations and antitumor activity compared to solvent-based paclitaxel[2][4]. This combination demonstrated improved progression-free survival and overall response rate compared to chemotherapy alone in phase III trials for advanced SCAC[3].
