Overview

**Retifanlimab + carboplatin + paclitaxel** is a combination regimen consisting of retifanlimab-dlwr (a programmed death receptor-1 (PD-1) blocking monoclonal antibody), carboplatin (a platinum-based chemotherapeutic agent), and paclitaxel (a microtubule inhibitor). This combination is FDA-approved for first-line treatment of adults with inoperable locally recurrent or metastatic squamous cell carcinoma of the anal canal (SCAC). Retifanlimab blocks the PD-1 receptor on T-cells, thereby restoring immune surveillance and anti-tumor immune response. Carboplatin induces DNA crosslinking and inhibits DNA synthesis, leading to apoptosis in rapidly dividing cells. Paclitaxel disrupts microtubule function, inhibiting cellular mitosis. The regimen demonstrated improved progression-free and overall survival compared to carboplatin/paclitaxel alone in clinical trials[1][3][5][7].
