Overview

**Retifanlimab + INCAGN02385** is a combination of two monoclonal antibodies under clinical investigation for cancer treatment. Retifanlimab is a humanized IgG4 monoclonal antibody that targets programmed cell death protein 1 (PD-1), blocking its interaction with ligands PD-L1 and PD-L2 to restore and enhance anti-tumor immune responses. INCAGN02385 is a monoclonal antibody antagonist of lymphocyte activation gene 3 (LAG-3), intended to inhibit a co-inhibitory immune checkpoint and further enhance T-cell mediated antitumor activity. The combination aims to address resistance mechanisms in patients with solid tumors, particularly recurrent/metastatic squamous cell carcinoma of the head and neck. The drugs are being evaluated as first-line therapy in PD-L1-positive, systemic therapy–naive patients, with ongoing phase 2 clinical trials assessing safety, optimal dosing, and efficacy[1][2][3][5][6].
